EliZyme™ ProofRead

EliZyme™ ProofRead polymerase is an effective and adaptable high-fidelity enzyme created for PCR applications that require greater sequence accuracy. With enhanced processivity and advanced buffer chemistry, it offers significant improvements in speed, yield, and sensitivity while also increasing the success rate of PCR with long and challenging templates.

This single enzyme is derived from Pfu DNA polymerase and has been engineered with proprietary mutations to improve DNA binding and increase processivity, resulting in shorter extension times (10-30 seconds per kb), higher yields, and the ability to amplify longer and more complex targets, including eukaryotic genomic templates over 17.5 kb in length.

EliZyme™ ProofRead polymerase exhibits superior accuracy and 3’-5’ exonuclease activity, which makes it 100 times more precise than Taq DNA polymerase, resulting in extremely low error rates. This makes it an ideal choice for applications requiring high accuracy such as cloning, site-directed mutagenesis, and sequencing. PCR products generated using this polymerase have blunt ends.

EliZyme™ ProofRead polymerase comes with an advanced buffer system that includes dNTPs, Mg, and enhancers, enabling high-fidelity PCR of a wide range of targets and fragment sizes with minimal or no optimization required. Additionally, it is available as a convenient 2X Ready MIX, with the option of a red dye for direct gel loading, saving time during reaction setup and analysis.
